The BD Accuri C6 benchtop flow cytometer from BD Biosciences (San Jose, CA) examines thousands of cells for reporter gene transfection efficiency and cell population heterogeneity. The system uses a blue and a red laser, two light-scatter detectors, and four fluorescence detectors with optical filters to detect fluorochromes such as fluorescein isothiocyanate (FITC), phycoerythrin (PE), peridinin chlorphyll protein (PerCP), and allophycocyanin (APC). It accepts additional reagents to screen cell populations, monitor DNA content, identify cells at different phases of the cell cycle, and examine heterogeneity and transient effects in reporter expression.

Screening thousands of cells for reporter gene expression levels is fundamental to understanding how genes are regulated inside the cell. The BD Accuri C6 can also measure the effects of gene knockdown when RNA interference (RNAi) is used to reduce gene expression. With multiparameter flow cytometry, one fluorescent antibody can be used to measure the intracellular level of the transfected siRNA target protein, while others can measure the cells' functional responses such as up- and down-regulation of surface markers or intracellular protein phosphorylation.
